Abstract:The antigenicity of Mycobacterium leprae (M. leprae)-derived cell membrane fraction was examined using human dendritic cells (DCs). Immature DCs internalized and processed the cell membrane components, and expressed M. leprae-derived antigens (Ags) on their surface. The expression of MHC class II, CD86, and CD83 Ags on DCs and CD40 ligand (L)-associated IL-12 p70 production from DCs were up-regulated by the membrane Ags. Moreover these stimulated DCs induced significantly higher level of interferon-gamma (IFN-gamma) production by autologous CD4(+) and CD8(+) T cells than those pulsed with equivalent doses of live M. leprae or its cytosol fraction. Both subsets of T cells from tuberculoid leprosy patients also produced several fold more IFN-gamma than those from normal individuals. Furthermore, the intracellular perforin production in CD8(+) T cells was up-regulated in an Ag-dose dependent manner. These results suggest that M. leprae membrane Ags might be useful as the vaccinating agents against leprosy. (AU).

Abstract:Peripheral blood lymphocytes from 105 subjects with different forms of leprosy and healthy contacts of leprosy patients weere stimulated in vitro different preparations of mycobacterial antugens alone or in combination with a suboptimal dose of phytohaemagglutinin (PHA). In nearly all individuals sonicated leprosy bacilli and PHA together gave a lower 3H-thymidine incorporation than didi the same dose of PHA alone. There was no difference in the degree of inhibition seen in the different patient groups or the healthy contacts. High doses of whole, washed Mycobacterium leprae, combined with PHA led to an increased thymidine incorporation in borderline tuberculoid leprosy patients who had experienced a reversal reaction, and in healthy contacts with less than 6 months exposure did not show an augmentation of the PHA-induced thymidine incorporation. The inhibition exerted by sonicated M. leprae was dose-dependent, seen even with very low doses of antigen, and was not due to direct Cytotoxicity, M. bovis, strain BCG, was weakly suppressive effect. There was no correlation between the suppressive effective of M. leprae antigens and the other mtcobacteria neither was there any correlation with the responses to the mycobacterial antigens alone. Many lepromatous leprosy patients showed significant suppression of background incorporation with addition of M. leprae antigens. This paper discusses wheter the apparent `non-responsiveness` in lepromatous leprosy could be due to active suppressor mechanisms operative in vitro.(AU).

Abstract:A hanseniase, doenca infecciosa cronica causada pelo Mycobacterium leprae, tem diferentes apresentacoes clinicas relacionadas a resistencia ou susceptibilidade ao bacilo. O quadro de linfocitos T de pacientes com hanseniase e artrite cronica possibilitou identificar e esclarecer os possiveis mecanismos imunologicos envolvidos na sua patogenese, atraves do estudo de 27 linhagens de celulas T envolvidos na resposta ao M. leprae, enfermos com hanseniase com artrite (n=4) e sem artrite (n=3). Para o esclarecimento da maioria dos eventos envolvidos no reconhecimento dos antigenos micobacterianos nos dois agrupamentos de doentes; foram investigados, a resposta a proteinas imunodominantes da micobacteria, e fenotipo das celulas envolvidas, o papel regulador destes linfocitos, atraves da determinacao das citocinas produzidas(AU).
